IGF-1 LR3: the hypoglycaemia risk is the part people skip

IGF-1 LR3 is discussed mostly in terms of muscle, and I want to talk about the first safety note, because I have spent twenty years managing hypoglycaemia and I do not think people are reading it properly.

IGF-1 has genuine insulin-like activity — it is in the name. The LR3 modification specifically evades the binding proteins that normally hold IGF-1 in check. So you have a molecule with insulin-like action, engineered to escape the buffering system that would otherwise limit how much of it is free and active.

Severe hypoglycaemia comes on faster than people expect, is genuinely frightening, and is a medical emergency. Somebody with no experience of it may not recognise it happening until they are past the point of managing it themselves.

Strongly agree, and the recognition point is the one I would underline.

If you have never had a hypo you do not know what it feels like from the inside, and the symptoms — confusion, irritability, sweating, shaking — include ones that impair the judgement you would need to act on them. That is what makes it different from most acute risks. The thing that goes wrong also degrades your ability to notice it going wrong.

Somebody alone, or asleep, has no second line.

And the second safety note is the longer-term one and is not a hedge either.

IGF-1 signals cells to divide. Raised IGF-1 has been associated in epidemiological work with higher rates of several cancers. Deliberately pushing it well above the normal range is not a small decision for anyone with a personal or family history of cancer.

The page calls this the most potent compound in its category and that is not a compliment. Potency cuts in every direction the molecule acts, and this one drives PI3K/Akt/mTOR — a pathway that is well characterised precisely because of how central it is to growth and to cancer biology.

The framing that lands for me: everything upstream in this category — the secretagogues, ipamorelin, CJC — asks the pituitary. This one skips the whole chain and applies the signal directly.

That is exactly why it is more potent, and exactly why the body's own regulation is not in the loop.

That is it precisely. The regulation you are bypassing is not an inefficiency. It is the safety system.

Also on the page and worth noticing: it is manufactured industrially as a laboratory reagent. Cell culture media.

Which means a substantial part of the supply exists because somebody needed it to grow cells in a flask, at a quality specification written for growing cells in a flask.
