Every discussion of frequency ends up being about half-life. I understand the definition — time for the concentration to halve — but I do not understand why it settles arguments the way people use it to.
Half-life keeps getting quoted at me. What does it actually tell me?

It settles fewer arguments than it is used to settle, and the gap is worth understanding.
Half-life tells you how long the compound is present. It does not tell you how long its effect lasts. Those come apart constantly:
- A compound can bind irreversibly and keep acting long after it has cleared.
- A compound can trigger a cascade that runs for days after a molecule that was gone in an hour.
- A compound can sit in plasma at a measurable concentration while doing nothing, because the concentration at the tissue that matters is a different number.
Plasma half-life is a pharmacokinetic measurement — where the drug is. What you care about is pharmacodynamic — what it does. They are related but they are not the same variable, and the whole reason both words exist is that people kept confusing them.
The other thing to ask is where the number came from.
Half-lives quoted for the unapproved compounds are frequently from rats, or from a structural analogue, or from one small study, and then get repeated until they sound like a physical constant. BPC-157 has confidently stated half-lives in circulation that trace back to remarkably little.
Compare that to something like semaglutide where the roughly one-week half-life is why it is a weekly injection, was measured properly in humans, and is the basis of an approved dosing schedule. That is a number doing real work.
And there is a species point buried in the rat ones. Small mammals clear most things faster than we do, so a rodent half-life is usually an underestimate for a human — sometimes by a lot, sometimes not, and which of those it is depends on the clearance route.
So even when the number is real it may not be a number about you.
So the honest summary is: half-life answers how long it is around, not how long it works, and for most of the compounds discussed here the figure itself is soft.
