A genuine trade-off rather than a settled question, so I want other views.
Freeze-thaw cycling damages peptides in solution. Ice crystal formation, concentration of solutes in the unfrozen fraction, pH shifts as buffer components crystallise out at different points. Each cycle costs something. So splitting into single-use aliquots and freezing those looks obviously right.
Against: every aliquot is an additional transfer, and every transfer is a contamination opportunity and a handling-loss opportunity. Doing it well needs a clean workspace and care.
On the number of cycles you would otherwise run, I think.
Two or three thaws over a short period — the aliquoting probably costs more than it saves. Ten thaws over months, it clearly saves more.
The other variable is who is doing it. Somebody with genuinely good aseptic technique gets the benefit. Somebody doing it on a kitchen table with the window open has multiplied their contamination opportunities by however many aliquots they made.
And whatever else, do not thaw and refreeze the same vial repeatedly. That is the case everybody agrees is bad and it is also the default behaviour if you do nothing.
Agreed on both. The honest answer is that it depends on the number of cycles and on your technique, and the number of cycles is the part you can actually estimate in advance.
Worth noting the fridge-door point from the storage post applies here too. If aliquots go in a freezer that gets opened constantly, the temperature cycling is doing some of the damage you aliquoted to avoid.
